A mutT derivative of a Bgl^- strain was plated on minimal glucose plates with 5-bromo-4-chloro-3-indolyl--D-glucoside. MMR^- sectors (e.g., mutH or mutS) are revealed by frequent blue Bgl⁺ papillae (demonstrating repeat tract instability). Here, an MMR^- mutant has risen at approximately the fifth cell division, resulting in a thin sector of the colony displaying repeat tract (or microsatellite) instability.
