Cover photograph (Copyright © 2001, American Society for Microbiology. All rights reserved.): Dal80p is a transcriptional repressor binding upstream of the genes it regulates. Up to 16 fluorescent foci occur in cells expressing GFP-DAL80, corresponding to the number of Saccharomyces cerevisiae chromosomes. The foci colocalize with 4',6'-diamidino-2-phenylindole staining and move through the cell cycle in the same way as DNA. Here, GFP-DAL80 is expressed in a bim100 mutant. Bim1p is required to correctly position the mitotic spindle. In bim1 mutants, the mitotic spindle misaligns perpendicular to its normal axis. As a result, green fluorescent protein-Dal80p "stained" material moves to opposite sides of the mother cell rather than spanning the neck between mother and daughter.
